SS-31 (also known as Elamipretide or MTP-131) is a synthetic tetrapeptide with an alternating aromatic-cationic sequence (D-Arg-dimethylTyr-Lys-Phe-NH2) that has been shown in cell studies to selectively accumulate in the inner mitochondrial membrane (IMM) through electrostatic interactions with cardiolipin, a phospholipid unique to the IMM.
Research using SS-31 has focused on its proposed ability to stabilize cardiolipin-cytochrome c interactions, which in turn supports electron transport chain (ETC) efficiency and reduces mitochondrial reactive oxygen species (ROS) generation in cell-based oxidative stress models. Rodent-model studies have used SS-31 as a pharmacological probe in cardiac ischemia-reperfusion injury models and age-related mitochondrial dysfunction paradigms.
